摘要 目的:建立测定血清中头孢呋辛(Cef)浓度的测定方法.方法:采用改进的HPLC法和YWGC18色谱柱(150mm×4.6mm),流动相为甲醇:醋酸-醋酸钠缓冲液(pH4.5)=(20:80),检测波长为274nm.结果:Cef浓度在(0.5~50)μg/ml范围内线性关系良好.日内、日间的RSD分别为2.39%、1.89%.结论:本法成本低、毒性小、操作简单快速、结果准确,适用于临床血清中Cef浓度的测定及药物动力学研究. AIM:To develop HPLC for the determination of cefuroxime.METHODS: Using a chemical bonded phase column(YWG C18 150mm×4. 6mm) with cefradine as the internal standard. The mobile phase was a mixture of methanol-NaAC buffer(20 : 80,pH = 4. 5) with a flow rate 1.0ml/min. The detected wave of cefuroxime was 274nm. RESULTS: Standard curve was dissolved in 0. 5 to 50μg/ml. The relative standard deviations of within-day and between-day were 2.39% (n = 5) and 1.89% (n = 4)respectively. CONCLUSION: This method is simple,fast and accurate.lt is used for determination of concentration of cefuroxime in blood and tissues and pharmacokinetic study clinically.
