摘要
目的观察羟基喜树碱(HCPT)对前列腺癌PC-3m细胞凋亡的影响,初步探讨其作用机制。方法采用四甲基偶氮唑盐(MTT)法、流式细胞仪(FCM)及激光共聚焦显微镜(CLSM)检测细胞增殖与凋亡变化。结果HCPT作用后PC-3m生长明显受抑,且呈时间-浓度依赖性;在FCM及CLSM上可见凋亡率增加。结论HCPT体外能有效抑制前列腺癌细胞株生长;但其作用机制目前尚需进一步研究。
Objective To study the feasibility of treatment of prostate cancer using HCPT and its mechanism. Methods Add a certain concentration of HCPT in human prostate cancer PC-3m cell incubated together, using the method of MTF,FCM and CLSM to detect the changes in cell proliferation and apoptosis. Results HCPT can inhibit the growth of human prostate cancer PC-3m cell obviously, and showing a time-a concentration-dependent;The rate of apoptosis increases in FCM and CLSM. Conclusion HCPT can inhibit the growth of human prostate cancer PC-3m cell in vitro tests. However, the mechanism is yet to be further studied.
