摘要
目的研究雷米普利对心肌梗死后心衰大鼠左心室重构及非梗死区胶原的影响。方法将结扎大鼠左冠状动脉前降支并饲养6周的24只存活大鼠随机分为假手术组(Sham group,n=8)、模型组(Model group,n=8)及雷米普利组(Ramipril group,n=8),连续灌胃给药4周后测定大鼠血流动力学参数,全心及左室肥厚指她(HW/BW,LVHW/BW),ELISA方法检测血清血管紧张素Ⅱ(AngⅡ)的含量,RT-PCR法测定非梗死区心肌组织胶原蛋白Ⅰ(col-Ⅰ)及胶原蛋白Ⅲ(col-Ⅲ)mRNA表达水平。结果雷米普利能明显升高左心室内压最大上升和最大下降速率(±dp/dtmax),降低左心室收缩压(LVSP)、左心室舒张末压(LVEDP)、心肌肥厚指数、血清血管紧张素Ⅱ的含量及下调col-Ⅰ、col-ⅢmR-NA表达水平(P<0.05-0.01),但对心率(HR)、收缩压(SBP)、舒张压(DBP)无明显影响(P>0.05)。结论雷米普利对心肌梗死后心衰大鼠心肌间质胶原重构有显著的抑制作用,抗心室重构的作用机制与下调胶原蛋白mRNA表达有关。
Objective To investigate the effects of Ramipril on left ventricular remodeling and collagen in noninfarction zone in heart failure rats after myocardial infaretion(MI). Methods The rat model of heart failure was established by left anterior descending coronary artery deligation. After 6 weeks, 24 surviving rats were divided into sham, model and ramipril groups at random, 8 rats in each, and were treated with medicines by intragastric administration respectively. At 10 weeks, we measuredleft ventricular end-dias- tolic pressure (LVEDP), left ventricular systolic pressure( LVSP), + dp/dtmax and-dp/dtmaxof left ventricular pressure, The content of Ang Ⅱ in serum was quantified with ELISA. The heart weight (HW)and left ventricular heart weight (LVHW)were weighed. HW/ BW and LVHW/BWwere calculated. Fttrthermore, the mRNA levels of col- I and col-Ⅲ were detected by RT-PCR in non-infarction zone. Results Ranfipfil decreased significantly LVSP and LVEDP, the content of Ang Ⅱ in serum, myocardial hypertrophy index, downregulated the mRNA expressiones of col- I and col- Ⅲ in non-infarction zone( P 〈 0.05 - 0.01 ), whereas increased + dp/dt- max( P 〈 0.05 - 0. 01 ). But no influence on HR, SBP and DBP( P 〉 0.05). Conclusion Ramiprit inhibit myocardial interstitial col- lagen reconstitution on heart failure after AMI in rats, which may be related to inhibiting the mRNA levels of col- I and col- Ⅲ during heart failure.
