Effect of Integrin Expression on Procollagen Synthesis of Fibroblasts from Scleroderma

摘要 目的 研究硬皮病成纤维细胞整合素表达对成纤维细胞合成原胶原的影响。方法 运用硫代修饰的整合素反义寡核苷酸阻断硬皮病成纤维细胞表面相应整合素亚基的表达 ,RT PCR检测整合素表达抑制后硬皮病成纤维细胞原胶原mRNA量的变化。结果 反义寡核苷酸能特异抑制硬皮病成纤维细胞相应整合素亚基的表达 ;整合素α5或 β1 亚基表达减少的成纤维细胞 ,原胶原proα1(Ⅰ )、proα1(Ⅲ )mRNA均显著降低。结论 降低硬皮病成纤维细胞整合素的表达 。 Objective Investigation of the effect of integrin on fibroblasts from scleroderma in the production of procollagen. Methods Phosphorothioate modified antisense oligonucleotides were used to interfere with the expression of integrin α 5 or β 1 subunit on scleroderma fibroblasts respectively, then the changes of procollagen mRNA were detected by RT PCR. Results The expression of integrin α 5 or β 1 subunit could be specifically inhibited by their antisense oligonucleotides respectively. Decreased expression of fibroblast integrin α 5 or β 1 subunit significantly lowered mRNA of procollagen α 1(Ⅰ) and α 1(Ⅲ). Conclusion Overproduction of procollagen may be inhibited in the level of transcription by lowering the expression of integrin on fibroblasts in scleroderma.
