摘要
目的观察L-盐酸赖氨酸(LMH)对脊髓损伤大鼠胰岛B细胞、血清胰岛素和血糖水平的影响。方法实验大鼠随机分为健康对照组、脊髓损伤对照组、LMH1组和LMH2组4组,无菌条件下对后3组大鼠行胸髓右侧半横断术,LMH1组和LMH2组术后分别立即按体质量621.5、310.8mg/kg予一次性腹腔注射LMH,脊髓损伤对照组注射等量生理盐水。48h后断头处死动物,观察LMH对脊髓损伤大鼠胰岛B细胞的免疫反应,检测血清胰岛素和血糖水平。结果与健康对照大鼠相比,脊髓损伤大鼠胰岛B细胞免疫组化反应强度明显减弱,血清胰岛素水平明显减低,血糖水平明显增加(均P〈0.01)。与脊髓损伤对照组相比,LMH1组(按体质量LMH621.5mg/kg)大鼠胰岛B细胞反应强度明显增强,血清胰岛素水平明显增加,血糖水平明显降低(均P〈0.05),而LMH2组(按体质量LMH310.8mg/kg)大鼠与脊髓损伤对照组相比上述指标差异无统计学意义(P〉0.05)。结论LMH可促进胰岛B细胞合成和分泌胰岛素而具明显降血糖作用。
Objective To observe the effect of L-lysine monohydrochloride (LMH) on pancreatic islet B cells, the levels of endogenous insulin and blood sugar levels in the injured spinal cord of rats. Methods The rats were randomly divided into four groups, the healthy control group, the spinal cord injury group, the LMH1 group and the LMH2 group. The rat model was produced by hemi-transection at the lower right thoracic spinal cord aseptically. Peritoneal injection of LMH was adopted among the rats in the LMH1 group and the LMH2 group at 621.5, 310.8 mg/kg respectively for once. Equivalent normal saline was injected into the rats of the spinal cord injury group. Results The immunohistochemical reactions of positive B cells were significantly weaker in the spinal cord injured group than in the control group (P〈0.01). The levels of endogenous insulin were significantly lower and the blood sugar levels were significantly higher in the spinal cord injured group than in the control group (P〈 0.01). The immunohistochemical reactions of positive B cells were significantly stronger in LMH group than in the spinal cord injured group (P〈0.05); the levels of endogenous insulin were significantly higher and the blood sugar levels were significantly lower in LMH1 group (621.5 mg/kg) than in the spinal cord injured group (P〈0.05). The immunohistoehemical reaction of positive B cells, the levels of endogenous insulin and the blood sugar levels had no significant difference in LMH2 group (310.8 mg/kg) compared with that in the spinal cord injured control group (P〉0.05). Conclusions The results indicate that LMH might participate in the regulation of B cells of pancreatic islet and thus the decrease of blood sugar level in the spinal cord injured rats.
