硒化壳聚糖与阿霉素联合及序贯给药对NB4细胞增殖的影响

摘要 目的研究硒化壳聚糖与ADM联合应用对NB4细胞增殖的影响,探讨两药序贯给药的不同效果。方法硒化壳聚糖(50,100mg/L)及ADM(0.5,1,2mg/L)同时或者序贯给药(先给予硒化壳聚糖后给予ADM或先给予ADM后给予硒化壳聚糖)作用于NB4细胞,MTT法检测药物对细胞增殖的影响;金氏公式评价两药的协同效果。结果硒化壳聚糖与ADM单独作用均可抑制NB4细胞增殖,二者联合应用呈协同作用;先给予硒化壳聚糖作用24h,再给ADM作用24h,对NB4细胞的抑制呈协同作用;先给予ADM作用24h,再给硒化壳聚糖作用24h,呈拮抗及单独相加作用。结论硒化壳聚糖与不同浓度的ADM(0.5-2mg/L)联合应用可协同抑制NB4细胞增殖;序贯应用硒化壳聚糖和ADM时,先给予硒化壳聚糖再给予ADM比先给予ADM再给予硒化壳聚糖协同效果好。 Objective To investigate the effects of selenium chitosan(Se) alone or combined with ADM on growth of NB4 cells and the efficacy of the different sequential administration. Methods MTT was used to determine the proliferative effects of the different sequential administration of Se and ADM on tumor cells. Jin' s formula was used to assess the synergistic effects of Se and ADM. Results Se or ADM inhibited NB4 cell proliferation in a dose-dependent manner, respectively. The combination of Se and ADM had synergis- tic effects on the inhibition of NB4 cell proliferation. The synergistic effect was observed after Se was first administered for 24 h and then ADM for another 24 h. However, simple addiction or antagonistic effect was observed after ADM was first administered for 24 h and then Se for another 24 h. Conclusion The combination of Se and ADM could synergistically inhibit NB4 cell growth, and the synergistic effects by sequential administration of Se then ADM is better than that by first use of ADM then Se.
