摘要
目的通过不同浓度氢氧化铝吸附Nogo-66后对SD大鼠免疫特征的影响,探索氢氧化铝佐剂对Nogo-66免疫原性的影响。方法氢氧化铝混悬液1ml与4%Nogo-66蛋白溶液以体积比例1∶1,1∶2,1∶3配制,充分微量震荡混匀30min。微板考马斯亮蓝蛋白定量法计算Nogo-66蛋白质的氢氧化铝吸附率。吸附氢氧化铝佐剂的Nogo-66蛋白免疫接种SD大鼠,免疫频次为1次/7d,共免疫4次。流式细胞术检测处于活跃分裂期细胞数目。结果单纯蛋白质组在所有实验组中,免疫原性最高,与其余实验组对比均具有统计学差异(P<0.05),中剂量佐剂组与小剂量佐剂组无统计学差异,二者与高剂量佐剂组对比均有显著性差异(P<0.05)。结论氢氧化铝佐剂对Nogo-66的影响表现为对免疫原性的抑制作用,其原因可能由于其刺激吞噬细胞活性,抑制抗原表位释放。
Objective To explore the effect of aluminum hydroxide(AH)on the immunogenicity of Nogo-66 in SD rats. Methods The mixtures of the aluminum hydroxide of 1 ml to 4% Nogo-66 were prepared in volume ratio of 1 : 1,1 : 2 and 1 : 3. The adsorption rate was calculated by Coomassie protein assay. The mixtures were immunized into SD rats once every 7 d for 4 times. The activated cell po- pulation during dividing phase by flow cytometry after the forth vaccination. Results The immunogenicity was higher in rats immu- nized with simple protein than that immunized with the mixtures ( P 〈 O. 05 ), and there was significant difference in immunogenicity after immunized with different ratios of the mixtures ( P 〈 O. 05 ). Conclusion The aluminum hydroxide could depress the immunoge- nicity of Nogo-66 by promoting the cytoactivity of phagocyte cells to inhibite the delivery of epitope.
