摘要 本文应用免疫吸附电镜法,对马铃薯、洋酸浆和曼陀罗植株中的马铃薯叶病毒(PLRV)粒体进行了检侧,其结果为:马铃薯茎、叶、幼芽、休眠块茎在铜网上每标准面积(SA=1000μm^2)中的病毒数量分别为:71364/SA,55909/SA,2273/SA,909/SA,洋酸浆茎和叶中的病毒数量分别为:2273/SA,15455/SA,曼陀罗茎中为:3636/SA,同时,当马铃薯茎和叶汁液稀释到1:32和1:8时,仍能明显地检测出PLRV粒体,证明免疫吸附电镜法对检测马铃薯植株中PLRV是一种快速、灵敏、直接、可靠的方法。 Potato leafroll virus in potato, Physalis floridana and Datura tatula were assayed by immunosorbent electron microscopy. Tests show:PLRV particle number per standard area(SA=1000μm^2)of grid in potato stem, leaf, sprout and tuber extracts are: 71364/SA, 55909/SA, 2273/SA, 909/SA respectively. PLRV partclcs number in stem and leaf of physalia floridana are about 2273/SA. PLRV particles can be detected in extracts at dilution of 1: 8 of potato leaves and 1: 32 of potato stem. Tests show that immunosorbant electron microscopy is a rapid, sensitive, direct and more reliable method for detection of PLRV in potato stems, leaves, sprouts, and it can be used as routine method for certification of seed potatoes. The diagnostic protocols for detection of PLRV in pctato stems and leaves have been suggested.
