摘要
目的研究miR-125b-1基因突变在人肺癌的发生发展中的意义。方法采用聚合酶链反应-单链构象多态性(polymerase chain reaction and single-strand conformation polymorphism,PCR-SSCP)分析法检测119例肺癌原发灶癌组织miR-125b-1基因突变情况。结果miR-125b-1在癌旁正常肺组织中未发现异常,在肺癌组织中存在基因突变,突变率34%(40/119),miR-125b-1基因突变与与癌的淋巴结转移及临床分期呈显著性相关,而与其它临床病理特征无关。miR-125b-1基因突变组肺癌患者的术后长期生存率显著低于高表达组(P<0.05)。结论肺癌组织中存在着miR-125b-1基因突变,miR-125b-1基因突变在肺癌的发生发展中可能起着重要作用。
Objective To investigate the role of miR-125b-1 gene in oncogenesis and progression of human lung cancer. Method PCR-SSCP was used to detect miR-125b-1 gene mutation in 119 specimen from primary site of lung cancer tissues and 42 samples of normal adjacent lung tissues. Result There were 40 cases of miR- 125b-1 gene mutation , and the analysis revealed that its mutation was positively correlated to the status of lymph node metastasis(P〈0.01) and clinical stage metastasis(P〈0.01), but no relationship was found between miR- 125b-1 mutation and other clinical features. The postoperative survival time in patients with miR-125b-1 gene mutation was significantly longer than those with low expression (P〈0.05). Conclusion There is miR-125b-1 gene mutation in lung cancer tissue, miR-125b-1 gene mutation probably plays a crucial role in lung carcinogenesis.
