摘要
采用痘病毒和牛痘病毒免疫莱航鸡制备卵黄抗体,分析了不同抗原浓度下抗体滴度的差异,以了解抗体产量受实验鸡生物节律影响的周期性变化。结果显示,在一定剂量窗范围内,免疫效果会随着抗原浓度的增加而加强。但过高的抗原剂量常会较低剂量引起免疫抑制。两次免疫之间,尤其是首次和二次免疫之间,间隔不宜过短,否则容易造成动物体内的免疫记忆,进而影响免疫效果。弗氏完全佐剂较弗氏非完全佐剂佐剂效力更强,但对机体组织伤害较大,两种佐剂先后交叉使用效果更好。卵黄抗体的产量在总体平稳中呈现明显的7 d和60 d生物波动。
In the study,Vaccinia virus and Cowpox virus were used to immune Leghorn chicken to prepare immunoglobulin yolk (IgY),the antibody titers of different immune dosage were analyzed,which could help recognize the change of antibody quantity was affected by biological rhythm of treated chicken. The results showed that an increase in antigen concentration may achieve higher antibody titers,but higher antigen level could also lead to immunosupression. Intervals between immunizations should be long enough to avoid immune memory. Freund's complete adjuvant was better than Freund's incomplete adjuvant,but which could make worse tissue damages,so the two adjvants should use alternately. Total IgY content showed markedly complex biological rhythm of 7 days and 60 days.
