18份大蒜种质资源遗传多样性的RAPD分析 被引量:11

摘要 应用RAPD标记对大蒜18个品种的遗传多样性和亲缘关系进行分析。从38个随机引物中筛选出6个多态性明显、反应稳定的引物,共扩增出36条谱带,其中多态性谱带27条。根据36个标记位点的信息,利用POPGENE32软件计算相关参数。结果表明,在物种水平上,多态性位点百分率P=75%,平均每个位点有效等位基因数Ne=1.4964,Nei s基因多样性指数H=0.2819,Shannon s多样性信息指数I=0.4158。根据Nei s遗传距离进行各类型间的UPGMA聚类分析,聚类结果将18个品种分为2大类群,成都二水早与其它地区大蒜种质资源有较大差别。 Genetic diversity and relationship of 18 samplers of Allium sativum L. germplasm resources were analyzed by randomly amplified polymorphic DNA ( RAPD ) markers in this paper. 6 RAPD primers were scveened from 38 arbitrary primers for genetic diversity analysis of garlic germplasm. Totally 36 bands were amplified, from which 27 bands exhibited polymorphic, thus the percentage of polymorphic loci is 75%. Based on the information of 36 loci and analyzed by POPGENE32 soft, the results indicated that , at species level, the average value of effective number of alleles Ne was equal to 1. 4964, Nei's gene polymorphic diversity index and Shannon's polymorphic information index were 0. 2819 and 0. 4158 respectively. According to Nei's genetic distance, the results of Unweighted Pairgroup Method Arithmetic Average (UPGMA) analysis showed that 18 samplers of garlic germplasm could be divided into 2 groups, and a distinctive germplasm, i. e. Ershuizao form Chengdu was significant different in the genetic relationship among the germplasm resources from another areas.
