摘要
通过正交实验筛选了一种用于紫丁香蘑原基诱导的合成培养基。合成培养基由复合碳源(木糖+糊精)、复合氨基酸氮源、无机盐、MS培养基中的微量元素及铁盐、生长因子、出菇刺激物等6部分构成,以珍珠岩为支持物。优化后培养基中含有5 g.L-1木糖、25 g.L-1糊精、0.1 g.L-1乙酸钠。在此培养基上,紫丁香蘑菌丝经24℃无光培养30d、再经16℃弱光刺激15 d后可产生83个原基。
In this paper a kind of synthetic medium of Lepista nuda for primordium induction was designed by orthogonal experiment. The culture medium was made from six parts as follows: the compound carbon source of xylose and dextrine, the nitrogen source of compound amino acids,inorganic salts, the microelement and the iron provided by culture medium MS, growth factors, mushroom stimulate things, etc. The halite was used to support medium. The optimized medium should contain 5 g·L^-1 xylose,25 g·L^-1 dextrine and 0. 1 g·L^-1 sodium acetate. When cultured in darkness for 30 d and then incited by weak light at 16℃ for 45 d in above synthetic medium,the Lepiat nuda mycelium could treat 80-83 or more of primordium.
