摘要 DNA等温扩增是在恒定温度下用非热变性的方法解开DNA双链的一种扩增技术。链置换反应指某些具有链置换活性的DNA聚合酶在延伸新链的同时将下游旧链剥离,它已被用于多种体外等温扩增技术,包括链置换扩增、滚环扩增、多重置换扩增、环介导的等温扩增等。其主要应用于DNA测序、全基因组扩增、基因芯片、微生物病原体检测等领域。 Isothermal amplification exploits strategies other than heating-denaturation to unwind double-stranded DNA.Strand displacement is such a process in which new DNA synthesis starts at a nick by displacing downstream strand while extention.Several strategies of DNA isothermal amplification,including strand displacement amplification,rolling circle amplification multiple displacement amplification,loop-mediated isothermal amplification,have been developed based on strand displacement reaction.And these methods have been applied in DNA sequencing, whole genome amplification, microarray fabrication and pathogen detection.
