核转录因子kappaB在急性心肌梗死的作用研究 被引量:2

摘要 目的探讨急性心肌梗死后(AMI)患者外周血单个核细胞(PBMCs)核因子-kappaB(NF-κB)活化与血浆肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)分泌的相关性。方法选取30例首次急性心肌梗死患者为观察组(AMI组),25例体检正常人为健康对照组,两组PBMCs的NF-κB表达应用细胞免疫化学染色检测,血浆细胞因子TNF-α、IL-1β由放射免疫法测定。对两组指标进行t检验及相关性分析。结果①AMI组血浆外周血PBMCs的NF-κB核染色阳性百分率、TNF-α、IL-1β分泌表达均显著增加高于健康对照组(P<0.01)。②AMI患者外周血PBMCs的NF-κB核染色阳性百分率与TNF-α、IL-1β表达均显著正相关(P<0.05)。结论AMI患者外周血PBMCs的NF-κB活性增高,可以通过促进细胞因子(TNF-α、IL-1β)的分泌来参与AMI的心源性休克、心力衰竭和心室重塑等病理生理过程。 Objective To investigate the role of NF - κB activation in the development of acute myocardial infarction, and to investigate the relationship of NF - κB activation on peripheral blood mononuclear cell (PBMCs) and the level of TNF - otand IL - 1β. Methods Thirty acute myocardial infarction patients defined as observing group (AMI group) ; twenty people defined as normal control group. And the activity of NF - κB was measured by immunocytochemical staining. Plasma cell cytokine ( TNF - ctand IL - 1β) were measured with radio immunity assay. Two sets index was matched with T - test and correlation analysis. Results Comparing with the normal control group, NF- κB activation, TNF- α and IL -1β was bonded to be increased in acute myocardial infarction (P 〈0.01 ). NF - κB activation were positively correlated with the level TNF - otand IL - 113(P 〈 0.05 ). Condution Increased NF - κB activation may be up regulate cytokine in acute myocardial infarction. NF - κB is one of factors for pathophysiology of cardiac shock, heart failure and ventricles remodeling in acute myocardial infarction.
