黄连炮制品全粉和水煎膏中生物碱类成分变化比较 被引量:3

Study on Alkaloids of Prepared Rhizoma Coptidis by Two Handling Methods

摘要 目的:考察黄连生、制品全粉和水煎膏中生物碱类成分的差异,探讨炮制辅料对生物碱类成分的影响。方法:将黄连炮制品分别制成全粉和水煎膏。高效毛细管电泳法测定生物碱相对峰面积。以苄基三乙基氯化铵为内标物,电泳条件:以50mmol.L-1Na2B4O7(H3PO4调pH=7.0)-MeOH(65∶35)溶液为背景电解质,运行电压15kV,紫外检测波长225nm,室温。结果:同一全粉样品中,各成分变化趋势相同,幅度基本恒定。顺序为:胆黄连>醋黄连>酒黄连>水黄连>姜黄连>生黄连>炒黄连>萸黄连>盐黄连。同一水煎膏样品中,各成分的变化趋势不同,不同样品中,相同成分,变化趋势也不同。同一样品全粉粉末和水煎膏比较,水煎后各成分均降低。但降低幅度不同。结论:辅料对黄连生物碱类成分影响很大,醋,胆汁,酒能促进其溶出,而姜汁,吴茱萸汁和盐水则起阻碍作用。样品不同处理方法,得出的结论不同,样品处理方式与研究目的关系密切。研究黄连炮制原理,宜选择全粉粉末为研究对象;研究药性,宜结合临床使用形式选择水煎煮作为样品处理方法。 Objective:To analyze the difference of alkaloid between the whole drug powder and the water extractive of prepared Rhizoma Coptidis(RC),and study the effect of excipients for fry to alkaloids.Methods:Friying with rice vinegar、with yellow rice wine and so on 8 different method were applied to manufacture RC,apply HPCE and regard benzyl tri-ethyl ammonium chloride as the internal standard substance to calculate the relative area.Results:the change trend of different component in the whole drug powder keep similar,and the range keep constant.The order was:RC fried with bile RC fried with rice vinegarRC fried with yellow rice wine RC fried with water RC fried with ginger juice RC without fried RC baked to yellowish RC fried with fructus evodiae juiceRC fried with saline solution.The change trend of various components in the same water extractive were different,and the change trend of the same component in the various samples was different.The assay of various components are decreased in the water extractive,but the trend is different.Conclusion:Excipients influent greatly to the alkaloid of RC,bile,vinegar and wine can promote it to dissolve,and ginger juice,fructus evodiae juice and saline solution can block it.The conclusion is different for various methods,and the methods for samples are related closely to the aim of study.Powder should be choiced as study object for studying the processed theory;the water extractive for studying the drug property.
