摘要
目的探讨银杏活脑胶囊对大鼠缺血性脑卒中微血管增殖和神经干细胞再生的影响。方法采用大脑中动脉栓塞模型,将雄性大鼠随机分成假手术组、缺血性卒中组和银杏活脑胶囊组。分组处理7 d后用5-bromo-2,-deoxyuridine(BrdU)免疫组化法观察海马齿状回(dentategyrus,DG)神经干细胞的增殖数;CD31免疫组化法观察梗死灶周围微血管增殖。结果假手术组大鼠海马DG区存在少量BrdU+细胞;银杏活脑胶囊组缺血侧齿状回颗粒细胞层边缘的颗粒细胞下层(Subgranular Zone,SGZ)的BrdU+细胞数和对侧BrdU+细胞数较缺血性卒中组有显著性增加(P<0.05);银杏活脑胶囊组梗死灶周围新生微血管数较缺血性卒中组有显著性增加(P<0.05)。结论银杏活脑胶囊治疗缺血性脑卒中的机制可能是促进缺血性脑卒中梗死灶周围微血管增殖和神经干细胞再生。
Objective to study effects of treatment with Yin Xing-Huo Nao capsule on the angiogenesis and neurogenesis in rat with cerebral ischemic.Methods Thirty rats are ramdomly divided into YinXing-Huo Nao group,cerebral ischemic group and sham operated group.On base of pattern cerebral occlusion.The YinXing-Huo Nao group is treated with oral YingXing-HuoNao capsule;sham operated group and the cerebral ischemic group are treated with 0.9% Nacl solution.Neurogenesis are detected by imrnunohistochemistry staining with Brdu and the Proliferation of cerebral microvessels were identified by immunostaining with antibodies to CD31 at 7 days.Results There are a small amount of neural stem cells and microvessels in DG and SGZ of rats in sham operated group;The numbers of neural stem cells and microvessels in YinXing-Huo Nao group are more than those in rats of cerebral ischemic group(P0.05).Conclusion Principle of therapeutic efficiency of YinXing-HuoNao on ischemic stroke is to increase microvessels and neural stem cells
