摘要
目的:探讨力竭运动后大鼠心肌肌钙蛋白-T(cTnT)分布及含量的变化。方法:将50只健康成年雄性SD大鼠随机分为一次力竭运动组和安静对照组,运动组采用游泳运动方式分别于力竭运动后即刻6、h、12 h、24 h取材,应用免疫荧光技术和图像分析方法对大鼠心肌肌钙蛋白-T分布及含量的改变进行研究。结果:力竭运动后各时相大鼠心肌cTnT分布变得紊乱,在心肌细胞间及细胞内的分布不均一;一次力竭后即刻、6 h和12 h右心室cTnT含量高于对照组(P<0.01)。结论:一次力竭运动均可使心肌cTnT分布变得紊乱,这可能是导致心肌机械功能异常的原因之一。
Objective: To observe the changes in the distribution and the content of troponin T(cTnT) in myocardial tissue of cats after exhaustion exercise at different time points. Methods: Fifty Male Sprague--Dawley rats were random- ly divided into sedentary control group and single bout of exhaustion exercise groups. Rats in the exhaustion exercise group were killed for the specimen of cTnT 0 h,6 h, 12 h and 24 h after exhaustion exercise. Immunofluorescence tech- nique and image--analysis were used to study the changes in distribution and content of cardiac troponin T(cTnT) in myo- cardial tissue of the rats. Results: After single bout of exhaustion exercise, myocardial cTnT distribution became disuni- form, in which myocardial cTnT became increased, decreased or even vanished in different sections. Right ventricle myocardial cTnT content 0 h,6 h and 12 h after single bout of exhaustion exercise was remarkable higher than that in the control group ( P 〈0.01). Conclusion': Single bout of exhaustion exercise may cause disorder and disuniformity in the distribution of myocardial cTnT, which may be one of the reasons for myocardial mechanical maifunction.
