摘要
对杜仲叶中绿原酸、总黄酮的大孔树脂分离工艺进行研究,并采用高效液相色谱对纯化产物进行检测,从9种大孔树脂中筛选出NKA-Ⅱ为最优树脂,最佳的吸附解吸条件为:上柱液pH值为4,流速1.0mL/min,用30%乙醇洗脱杜仲叶中绿原酸,50%~70%乙醇洗脱杜仲叶中黄酮类物质。杜仲叶粗提物依次经过NKA-Ⅱ大孔树脂、聚酰胺树脂和SephadexLH-20树脂的分离纯化,最终得到4种组分,经高效液相色谱检测分析,可能为绿原酸、金丝桃苷或陆地锦苷、槲皮素-乙酰糖苷和槲皮素。
The technology that using macroporous resins to isolate the chlorogenic acid and total flavone from Eucommia ulmoidues leaves was studied and the purified compounds were identified by high performance liquid chromatography (HPLC). The NKA- Ⅱ column was selected as the optimal resin among nine kinds of macroporous resin and the optimum conditions for adsorption and desorption were as follows: pH value of sample solution was 4, the flow velocity was 1.0 mL/min, suitable desorption solvent to NKA-Ⅱ were 30% ethanol for chloregenic acid and 50%-70% ethanol for flavone. The crude extracts of Eucommia ulmoidues leaves were separated and purified by NKA- Ⅱ macroporous resin, polyamide resin and Sephadex LH-20 resin, respectively, obtained four components in the end. These four components identified by HPLC, the results showed that there could be chlorogenie acid, hyperoside or hirsutin, quercetin-acetyl-glucoside and quercetin.
