摘要 目的建立以去甲万古霉素为内标的人血清中万古霉素HPLC测定法。方法血样处理采用10%三氯醋酸沉淀法,色谱分析采用Agi-lent C18(250mm×4.6mm,5μm)分析柱,流动相为甲醇-乙腈-0.05mol.L-1磷酸二氢钾溶液(15:2:83,pH3.2),紫外检测波长236nm,流速1.0mL.min-1,柱温35℃。结果万古霉素的线性范围为1.3mg.L-1~83.2mg.L-1(r>0.999);平均回收率为100.6%;日内、日间RSD均<4%。结论此法具有采血量少,检测快速、稳定、灵敏度高而成本低特点,适合临床常规血药浓度的监测。 OBJECTIVE To establish an high performance liquid chromatography (HPLC) assay for the determination of concerttration of vancomycin in human plasma using norvancomycin as internal standard.METHODS The pretreatment of the blood samples was simply sedimentation with 10% trichlomacetic acid.Then the samplewas separated on Agilent C18 column with a mixture of methanol-acetonitrile-0.05mol·L-1 potassium dihydrogenp hosphate(15∶2∶83,pH3.2).The UV wavelengthw as 236nm.The flow rate was 1.0 mL·min-1.The temperature of the column was 35℃.RESULTS A linearity was obtained from 1.3mg to 83.2mg·L-1 of vancomycin with good correlation(r〉0.999).The average recovery was 100.6%.The RSD for intra-day and inter-day assays were less than 4%.CONCLUSION It appears to be a rapid,convenient and accurate method for determination of vancomycin in plasma.
