摘要 目的:检测蛋白激酶D1(PKD1)在胃癌细胞中的表达,并探讨其对胃癌细胞增殖侵袭的影响。方法:体外培养胃癌细胞系BGC-823,使用PKD1甲基化特异性阻断剂5-杂氮-2′-脱氧胞苷(5-Aza-CdR)处理胃癌细胞,Western Blot法检测胃癌细胞中PKD1蛋白的表达情况,MTT法检测胃癌细胞的增殖,Transwell实验检测胃癌细胞的侵袭力。结果:5-Aza-CdR作用后,胃癌细胞中PKD1蛋白表达明显增加(P<0.05),细胞增殖速度减慢,侵袭能力减弱(P<0.05)。结论:胃癌细胞中PKD1表达的下调在胃癌的发病过程中可能起重要作用,并可能与胃癌细胞增殖侵袭能力改变有关。 Objective To investigate the expression of PKD1 in the gastric carcinoma cells,and to explore its influence on the proliferation and invasion.Methods Gastric carcinoma cell lines BGC-823 was cultured in vitro and treated with DNA methylation specific inhibitor 5-Aza-CdR.The expression change of PKD1 protein and proliferation of BGC-823 cells were detected with western-blot and MTT,respectively.The invasiveness was detected with Transwell assay.Results The expression of PKD1 in BGC-823 cells was significantly increased after treating with 5-Aza-CdR(P0.05),and the proliferation and invasiveness were significantly decreased(P0.05).Conclusion The down-regulation of PKD1 expression may play an important role in the pathogenesis of gastric carcinoma,and it may be related with the changes of proliferation and invasion abilities in gastric carcinoma cells.
