摘要
用酚/氯仿提取法提取了蓝鳃太阳鱼、绿色太阳鱼肌肉组织的总DNA,利用特异引物进行PCR扩增,得到1 119bp Cyt b基因片段,纯化后送上海生工测序。所得序列用Mega3.1软件分析,并结合GenBank中其余9种太阳鲈属鱼类Cyt b基因序列用NJ法构建太阳鲈属鱼类系统发育树。结果表明:(1)蓝鳃太阳鱼与绿色太阳鱼序列相似度为82.3%,变异位点198个,平均转换/颠换比为2.1;(2)蓝鳃太阳鱼与L.humilis亲缘关系较近,绿色太阳鱼与L.symmetricus亲缘关系较近,蓝鳃太阳鱼与绿色太阳鱼遗传距离为0.206。
The DNA sequences of 1 119 bp mitochondrial cytochrome b gene fragment ofLepomis macrochirus and Lepomis cyanellus were analyzed with Mega 3.1. The molecular phylogenetic tree was based on the sequences of 11 Lepomis species with neighhor-joiningmethod (NJ) respectively. The result were as follows:(1) The sequences identity matrix got 82.3%, and the substituted position between the two sequences got 198, with the transition/transversion ratio of 2.1.(2)There were cluster relationships between Lepomis macrochirus and L. humilis, Lepomis cyanellus and L. symmetricuz respectively. The genetic distance between Lepomis macrochirus and Lepomis cyanellus got 0.206.
