摘要 以鼠尾藻为原料,采用酶与化学结合的方法提取膳食纤维,通过酶用量、酶解时间、酶解温度、氢氧化钠浓度、氢氧化钠提取温度、氢氧化钠用量、氢氧化钠提取时间7个单因素试验和2个正交试验,研究确立了提取鼠尾藻膳食纤维的最佳工艺条件。结果表明:提取膳食纤维的最佳工艺条件为在40℃条件下用30:1的蛋白酶与纤维素酶酶解2 h,再用20倍1.5%NaOH溶液在55℃提取1 h,膳食纤维的产率可达15%左右,颜色为米黄色。 The technique to extract dietary fiber from Sargassum thunbergii with the combination of enzymatic and chemical methods was studied. The each of seven factors that could affect the efficiency of extracting dietary fiber was evaluated by two orthogonal test. The seven factors were the amount of used enzyme, the time and temperature of enzymatic digestion, the concentration of NaOH, the amount of added NaOH, and the time and temperature of extracting reaction with NaOH. An optimum condition to extract dietary fiber from Sargassum thunbergii was established. It is that the material was first hydrolyzed with a mixture of protease and cellulose (the ratio of two enzymes is 30:1) at 40 ℃ for 2 h and then a 20 volumes of 1.5% NaOH solution was added and the reaction was kept at 55 ℃ for 1 h. Under this condition, the yield of dietary fiber reached 15% and the fiber had a light yellow color.
