摘要
通过菌种复配研究了从黄姜中提取皂素的一种环境友好的生物方法。首先从生长黄姜的土壤和常见霉菌中筛选并驯化出三株菌,一株产纤维素酶活力较高的T.reesei,一株产β-糖苷酶活力较高的A.niger和一株产木聚糖酶活力较高的A.oryzae。其次研究了这三株菌四种复配模式发酵体系中酶活力和皂素得率的变化。与纯种发酵相比,A.niger与T.reesei复合发酵对提高体系中酶活和皂素得率有协同促进作用。最后在此基础上,进一步优化了这两株菌的发酵顺序。当T.reesei发酵第3d时投加A.niger继续发酵6d,皂素得率最高,可达到78.25%。
An environmental-friendly biotechnological method for the production of diosgenin from Dioscorea Zingiberensis C.H.Wright tubers by mixed culture was proposed in this study.Three fungal cultures,Trichoderma reesei,a good producer of cellulose,Aspergillus niger,a high β-glucosidase and Aspergillus oryzae,a high level of xylanase,were selected by a preliminary screening and acclimating from soil isolates and other available fungal cultures.The effects of four combinations of the three strains on enzyme activities and diosgenin yield were studied.The result showed that compared with pure culture fermentation,the combination of Aspergillus niger and Trichoderma reesei had a synergitic effect on enzyme production and diosgenin accumulation.The fermentation mode was optimized.the highest diosgenin yield of 78.25% was obtained when Aspergillus niger was fermented for 6 days followed by fermentation of Trichoderma reesei for 3 days.
