不同方法精制的磷脂酰胆碱纯度及脂肪酸组成分析 被引量:2

ANALYSIS OF PURITY AND FATTY ACID COMPOSITION OF PHOSPHATIDYLCHOLINE REFINED BY DIFFERENT METHODS

摘要 利用高效液相色谱测定了不同精制提纯方法制备的磷脂酰胆碱试样的纯度.结果表明:提取的方法不同,磷脂酰胆碱的纯度也不同,且存在较大的差异,磷脂酰胆碱纯度高低顺序为柱层析法纯化的磷酯>冷冻法纯化的磷酯>乙醇溶剂法萃取的磷酯>粉末磷脂>浓缩磷脂.利用气相色谱分析了各磷脂酰胆碱试样的脂肪酸组成.结果显示:主要含棕榈酸、硬脂酸、油酸、亚油酸及亚麻酸等5种脂肪酸,亚油酸、亚麻酸等不饱和脂肪酸与磷脂酰胆碱纯度呈现正相关性,硬脂酸等饱和脂肪酸与磷脂酰胆碱纯度呈现负相关性,油酸的变化趋势不明显.建立了亚油酸、棕榈酸、不饱和脂肪酸、人体必需脂肪酸等与磷脂酰胆碱纯度的关系曲线,其线性关系良好. The purities of five phosphatidylcholine samples refined by different methods were analyzed by HPLC. The results show that the purities of phosphatidyleholine samples refined by different methods differ from each other and have significant difference. The purities of the phosphatidylcholine samples were as fol- lows: from high to low, phosphatidyleholine purified by column chromatography, phosphatidyleholine purified by freezing process, phosphatidylcholine purified by ethanol extraction, powdered phosphatide and concentrated soybean phospholipid. Moreover, gas chromatography was used to analyze the fatty acid compositions of these phosphatidyleholine samples. The results show that phosphatidyleholine mainly contains five fatty acids, such as palmitie acid, stearic acid, oleic acid, linoleic acid and linolenic acid; the contents of unsaturated fatty acids, such as linoleic acid and linolenic acid, show positive correlation with the purity of phosphatidylcholine; the contents of saturated fatty acids, such as palmitic acid and stearic acid, show negative correlation; and no obvious change occurs for oleic acid. Furthermore, the linear relationship curves with good linearity between the contents of linoleic acid, palmitic acid, unsaturated fatty acid and essential fatty acid and the purity of phosphatidyleholine were established respectively.
