摘要
目的:研究缺氧诱导因子-1α(HIF-1α)与基质金属蛋白酶-2(MMP-2)在胃低分化腺癌中的表达,并探讨二者在胃低分化腺癌浸润转移过程中的相互关系及作用。方法:采用S-P免疫组化方法,对胃低分化腺癌石蜡标本54例及正常胃组织20例,每例切片两张,分别检测HIF-1α和MMP-2的表达情况,结果进行统计学处理。结果:胃低分化腺癌组织中HIF-1α、MMP-2阳性表达明显高于正常胃组织(P<0.05),二者与胃低分化腺癌的浸润深度、淋巴转移、TNM分期均显著相关(P<0.05),HIF-1α与MMP-2的表达呈正相关(r=0.580,P<0.05)。结论:HIF-1α和MMP-2在胃低分化腺癌组织中的表达呈正相关(r=0.580,P<0.05),表明二者在胃低分化腺癌的侵袭转移过程中存在一定的协同作用。二者可能成为判断预后的指标。
Objective:To investigate the expression of hypoxia inducible factor-1α (HIF-1α) and matrix metalloproteinase-2 (MMP-2) in poorly differentiated gastric adenocarcinoma (PDGA) and their relationship with tumor cell invasion and metastasis. Methods:We detected the expression of HIF-1α and MMP-2 in the paraffin-embedded specimen of 54 cases of poorly differentiated gastric adenoearcinoma (two slices for one case) and 20 cases of normal gastric tissue by immunohistochemistry. The results were analyzed statistically. Results:The positive expression rates of HIF-10. and MMP-2 were significantly higher in PDGA than those in the normal gastric tissue,with a significant difference between the two (P 〈0.05) . HIF-1α and MMP-2 was obviously correlated with the depth of invasion, lymph node metastasis and TNM stage of PDGA (P 〈 0.05), and there was a positive correlation between HIF- 1α expression and MMP- 2 expression (r= 0. 580, P〈 0.05). Conclusion :A positive correlation between the HIF-1α. expression and MMP-2 expression in PDGA suggests both have a certain synergy during the invasion and metastasis of PDGA and both could be indicators of prognosis.
