针刺百会和大椎穴对帕金森病小鼠多巴胺能神经元的保护作用 被引量:14

摘要 目的探讨针刺百会和大椎穴对帕金森病(PD)小鼠多巴胺能神经元的保护作用及可能机制。方法C57BL/6雄性小鼠随机分为5组:正常组、模型组、针刺组、美多巴组、针刺结合美多巴组(针美组)。后4组首先腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)建立PD动物模型,后3组造模后采取不同的治疗措施,造模后动物存活14d。采用爬杆法测量小鼠行为学改变,应用高效液相电化学内标法测定前脑单胺类神经递质含量,用免疫组织化学技术检测黑质酪氨酸羟化酶(TH)阳性神经元数目。结果针刺、针美组小鼠爬杆时间较模型组短(P<0.05)。与正常组比较,各组动物前脑内多巴胺及其代谢产物3,4-二羟基苯乙酸含量明显降低,3个治疗组与模型组比较虽然无明显差异,但从数值上看降低的程度较模型组低。造模后14d,模型组TH阳性神经元数目较正常组减少(P<0.01),3个治疗组也较正常组减少(P<0.01),但较模型组增多(P<0.05)。结论针刺百会、大椎穴或针刺结合美多巴可改善PD小鼠的行为障碍,这种作用可能与减少多巴胺能神经元的丢失有关。 Objective To study the protection effect of acupuncture of Baihui and Dazhui points on the dopaminergic neurons in Parkinson's disease(PD) mice and explore the possible mechanism of acupuncture in treating Parkinson's disease.Methods Thirty male C57BL/6 mice were randomly divided into five groups:normal control group,model group(PD model was made by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP),acupuncture group,Medopa group,acupuncture mixed with Medopa group(six mice for each group).The animals were killed on the 14th day after first injectiion of MPTP.The changes of behavior of rats were detected by climbing rod method.The levels of forebrain monoamines neurotransmitters were measured by high pressure liquid chromatograph.The TH+(Tyrosine hydroxylase) dopaminergic neurons in the substantia nigra were counted in every group by immunohistochemistry technique.Results The time of climbing rod was shorter in acupuncture group and acupuncture mixed with Medopa group(P〈 0.05) than in model group.Compared with normal control group,the levels of dopamine(DA)、dihydroxv-phenyl acetic acid(DOPAC) were lower in all other groups(P〈0.05).Meanwhile,the number of TH+ positive neurons was less in model group than in normal group,and also less in 3 treatment groups than in normal group,but more in 3 treatment groups than in model group.Conclusions Acupuncture of Baihui and Dazhui points can improve PD mice's behavior disorders,which is probably related to reducing the dopaminergic neurons missing.
