TGF-β_1抑制人类膀胱逼尿肌细胞中缝隙连接蛋白-43表达的实验研究

摘要 目的研究TGF-β1对人类膀胱逼尿肌细胞(hBSMCs)中缝隙连接蛋白-43(Cx-43)表达的影响。方法进行hBSMCs的原代培养(取材于膀胱全切患者的膀胱组织),用人类重组TGF-β1对hBSMCs进行干预后,利用免疫细胞化学和Western-blot-ting检测Cx-43在hBSMCs中的表达情况。结果TGF-β1干预后,Cx-43的免疫反应性显著降低(P<0.0001),同时Cx-43蛋白的表达显著下调(P<0.05),Cx-43的磷酸化作用降低。结论TGF-β1抑制人类膀胱逼尿肌细胞中Cx-43的表达。 Objective To examine the inhibitive effects of TGF-β1 effects on Cx-43 expression in cultured human detrusor smooth muscle cells (hBSMCs). Methods hBSMCs cultures established from patients undergoing cystectomy were treated with recombinant human TGF-β1. Cx-43 expression was then examined by Western blotting and immunocytochemistry. Resuits Stimulation with TGF-β1 led to significant reduction of Cx-43 immunoreactivity. Cx-43 protein expression was significantly downregulated. Interestingly, low phosphorylation species of Cx-43 were particularly affected. Conclusion This experiments demonstrated a significant downregulation of Cx-43 by TGF-β1 in cultured human bladder smooth muscle cells.
