低KAI1 mRNA水平与胰腺癌的浸润转移有关

Reduced KAI1 gene level is associated with advanced stage of primary pancreatic cancer

目的为了解ＫＡＩ１ｍＲＮＡ水平与胰腺癌的浸润转移关系。方法用Ｎｏｒｔｈｅｒｎｂｌｏｔ法对１２例正常胰腺组织和３０例原发性胰腺癌组织中ＫＡＩ１ｍＲＮＡ基因的表达进行检测分析。结果正常胰腺组织中该基因的表达水平呈阴性或弱阳性。相比之下，该基因在８３．４％（２５／３０）胰腺癌组织中有稳定的表达水平。在早期胰腺癌中出现的高表达ＫＡＩ１基因水平，与进展期伴有淋巴结及远处转移的晚期原发性胰腺癌比较。差异有非常显著性（Ｐ＜０．０１）。在低分化的胰腺癌中该基因的翻译水平显著高于高中分化的胰腺癌（Ｐ＜０．０５）。该基因与胰腺癌患者的性别、年龄和肿瘤的大小无关。结论高调节的ＫＡＩ１基因出现在早期胰腺癌中，而低表达的ＫＡＩ１基因可能与胰腺癌的浸润转移有密切关系。

Objective To determine the relationship between KAI1 gene level and advanced stage of primary pancreatic cancer. Method KAI1 mRNA expression was studied with Northern blot analyais in 12 normal pancreas and 30 primary pancreatic cancers. Results Weak or negative signals were found in normal pancreas and stable-expressed signals in 83.4 % (25/30) pancreatic cancer specimens. The KAI1 mRNA levels were significantly higher in earlier stage of pancreatic cancers as compared with advanced stage with lymph node or distant metastases (P< 0.01). Well or moderately differentiated tumors had significantly lower KAI1 mRNA expression than poorly differentiated cases (P< 0.06). Conclusion It was suggested that down regulation of KAI1 gene might have a role in metastasis of pancreatic cancer as it was high in its early stage. [
