胰岛素、尼莫地平对早期大鼠脑缺血再灌注神经细胞凋亡的影响

Effects of Nimotop and Insulin on Apoptosis of Neuronal Cell in Early Period of Focal Brain Ischemia Reperfusion Injury in Rats

摘要 目的:研究胰岛素、尼莫地平单药及联合用药对脑缺血再灌注时神经细胞凋亡的影响。方法:建立SD大鼠脑缺血再灌注模型,将40只SD大鼠随机分为5组,分组使用不同药物并进行对照,采用原位末端凋亡荧光法检测凋亡细胞。结果:与对照组相比,用药各组的细胞凋亡数明显减少,差异有显著性(P<0.05)。联合用药组与单独给药组差异具有显著性(P<0.05)。结论:胰岛素、尼莫地平联合用药可以明显抑制脑缺血再灌注时细胞凋亡的发生,具有较好的神经保护作用。 Objective: To study the effect of insulin, nimotop and their combination on neuronal apoptosis of focal brain ischemia reperfusion injury in rats. Method: 40 SD rats were divided randomly into five groups: sham operative group(O group), control group (A group), insulin group (B group), nimotop group (C group) and insulin-nimotop group (D group). Experimental models were established by intraluminal occlusion of the middle cerebral artery in rats. Apoptotic cells were detected by TUNEL technique. Results: Compared with the control group, the number of apoptotic cells in other groups were reduced evidently(P〈0.05, especially in insulin-nimotop group. Conclusion: Both nimotop and insulin can reduce apoptosis occurrence, but this effect can be enhanced when they are applied together. So making use of insulin and nimotop after cerebral ischemia can play a role in neuroprotection.
