精核蛋白与男性不育

Relationship of Protamine and Male Infertility

目的：探讨精子中精核蛋白含量与男性不育的关系、方法：从92例不育男性和16例正常生育男性的精子中提取精核蛋白.在醋酸-尿素聚丙烯酰胺凝胶（acid-ureaPAGE，pH4.7）中电泳.经薄层扫描，测得各蛋白区带（组蛋白和精核蛋白）的相对含量，分析并计算各蛋白区带的相对比值。结果：9例病人的精核蛋白完全缺乏。27例妻子流产男性精子畸形率升高明显（P＜0.01）、HP1～3／TBP降低（P＜0.01）。精子畸形率与精核蛋白的转化率成负相关。与正常生育组相比，92例男性不育症的总组蛋白（TH）与总碱性蛋白（TBP.包括TH、HP1、HP2、HP3）的比值增高;精核蛋白的含量由正常86.6±5.1％减少为64.1±14.4％（P＜0.01）;精子密度、精子活动力、畸形率、HP1／TBP、HP2＋3／HP1等相差显著（P＜0.01）。结论：精核蛋白含量异常的精子与畸形率、活动力、密度以及不明原因的流产等都具有相关性。

Objectives:To study the relation between content of nuclear basic protein and male infertlilty. Methods:Sperm basic nuclear protein was extracted from 92 infertile men and 16 fertile men. The acid soluble protein was eletrophoresed in 12. 5% acid--urea polyacrylamide gels electrophoresis (PAGE ). The relative proportions of histones and protamines were determined by microdensity scanning and ratio of basic nuclear protein was analysed. Results:The results showed that protamine was completely absent in 9 cases. The morphological abnormality of sperm in abortion group was more predominant higher than that in control group (P<0.01 ) while HP1 -- 3/TBP was decreased (P<0.01). The transformation rate of nuclear basic protein was negatively correlated with the morphological abnormality of sperm (r=-0. 79, P<0.01 ). The content of protamine decreased from 86. 6±5. 1% to 64. 1 ±14. 1 %. The quantity and motility, viability teratogenesis of sperm, HP1/TBP, HP2+ 3/HP1 were different significantly. Conclusions: The decrease of protamine is related to sperm teratogenesis,motility,viability and unexplained abortion.
