C-FLIP基因在甲基硫代腺苷诱导结肠癌RKO细胞凋亡过程中的作用

摘要 目的:探讨甲基硫代腺苷(MTA)对结肠癌RKO细胞凋亡的作用及其相关机制的研究.方法:采用Hoechst33258染色技术检测MTA对结肠癌RKO细胞凋亡的影响,并采用Realtime PCR和Western Blot的方法在mRNA和蛋白水平研究cFLIP基因在MTA诱导的结肠癌RKO细胞凋亡过程中的作用.结果:MTA对结肠癌RKO细胞具有明显促进凋亡的作用.在这一过程中,cFLIP基因在mRNA和蛋白水平上均出现明显下调情况.结论:MTA对RKO细胞具有明显凋亡诱导作用,cFLIP基因的下调可能起到了重要的作用. AIM:To study the effects and its mechnism of methylthioadenosine on apoptosis in RKO cells.METHODS:Hoechst33258 staining was used to detect the effects of methylthioadenosine on apoptosis in RKO cells,and the mRNA and protein level of cFLIP in this course was detected with the mothod of Realtime PCR and Western Blot.RESULTS:Methylthioadenosine could significantly induce apoptosis of RKO,and mRNA and protein level of cFLIP were downregulated significantly in this course.CONCLUSION:Methylthioadenosine could induce apoptosis in RKO cells,and the downregulation of cFLIP gene might play a major role.
