摘要
为研究柔嫩艾美耳球虫感染雏鸡后,雏鸡血清IgG含量的变化,用8×105个柔嫩艾美耳球虫卵囊感染10日龄雏鸡,用饱和硫酸铵盐析法分别提取感染前和感染后第1,2,3,4,5,6,7,8,12,15天鸡血清中的IgG,Sephadex G-25凝胶过滤层析,SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE),对纯化的IgG做纯度鉴定,结果表明,盐析产品纯度较高。通过紫外吸收法测定其含量,感染前IgG含量是(1.07±0.05)g/L,对照组感染后第4天(14日龄)达到峰值(1.90±0.07)g/L,感染组在感染后第6天(16日龄)达到最高值(2.31±0.07)g/L,感染后第12天(22日龄)对照组和感染组降到最低值,分别是(1.35±0.17)g/L和(1.32±0.05)g/L,感染后第5天(15日龄)差异显著(P<0.05),第6,7天(16,17日龄)两组差异极显著(P<0.01),其它天差异不显著(P>0.05)。
An experiment was conducted to study the dynamic changes of the Content of IgG in the serum of coccidian. Ten-day old chickens were infected with 8 × 10^5 sporulated oocysts. SDS Polyacrylanmide Gel Electrophoresis (SDS-PAGE) demonstrated that the serum IgG that was isolated by ammonium sulfate followed by SephadexG-25 gel filtration was pure. The results suggested that the content of IgG in serum before infection was(1.07±0.05)g/L,and it reached the highest point on the 4th day after being infected in control group (1.90±0.07)g/L,while on the 6th day in infection group it was (4.82±0.42)g/L. After the 12th day, the content of IgG of control group and infection group respectively reached the lowest point(1.35±0.17) g/ L and(1.32±0.05)g/L. The content of IgG in infection group serum was significantly different (P〈0.05) on the 5th day,and on the 6th and 7th days the difference was highly significant (P〈0. 01) from the control group. On the other days the difference was not significant.
