摘要
目的探讨未成熟心肌缺血再灌注损伤及三磷酸腺苷-氯化镁(ATP-MgCl2)的保护作用。方法采用3~4周龄兔心建成离体左心做功模型,实验动物分为低温组:局部单纯低温组(10~15℃);Thomas组:低温加Thomas液灌注组;ATP-MgCl2组:低温加ATP-MgCl2心停搏液灌注等3组,各组12只,均全心停循环90分钟,复灌流60分钟。记录其心功能恢复率及病理生理的改变。结果ATP-MgCl2组与低温组和Thomas组比较,心功能和心肌游离核苷酸含量恢复较好,冠状静脉流量高,心肌超微结构改变轻,而其磷酸肌酸激酶,心肌含水量及心肌线粒体丙二醛含量较低。结论ATP-MgCl2作为心停搏液添加剂。
Objective To evaluate the injury of ischemia reperfusion to immature rabbit heart and the protective effects of ATP MgCl 2 cardioplegia. Methods The isolated left ventrical working heart model was used from 3-4 weeks old rabbit heart. Thirty six rabbits were studied in three groups. Topical cooling along (10-15℃), single dose St Thomas cardioplegia plus topical cooling, single dose ATP MgCl 2 cardioplegia plus topical cooling. The hearts were subjected to 90 min of global ischemia and followed by 60 min of reperfusion, and the recovery of heart function and physiopathology changes were measured. Results The recovery of cardiac function and myocardial nucleotides content were good, the coronary flow was high and the ultrastructial changes of myocardium were slight, but the myocardial water content, molodiadehyde (MDA) of myocardial mitochondrial and creation phophate kinase (CPK) leakage of coronary effluent were lower in ATP MgCl 2 than those of either topical cooling and St Thomas groups. Conclusion The results suggest that as a valuable addition to hypothermic cardioplegia, ATP MgCl 2 produces better protection of the myocardium in immature rabbit heart.
