摘要 目的在Chang Liver细胞中观察黄芩苷处理对CYP3A4、CYP2C9和CYP2C19表达的影响。方法在Chang Liver细胞中分别加入DMSO0.1%(v/v)(溶剂对照)和不同浓度的黄芩苷(0.01、0.1和1μM)24、36和48h后,用半定量RT-PCR的方法分别检测CYP3A4、CYP2C9和CYP2C19表达量的改变。结果用不同浓度黄芩苷分别处理Chang Liver细胞24和36h,CYP3A4、CYP2C9和CYP2C19的表达均呈现出增强的趋势;但是黄芩苷处理48h后各浓度对CYP3A4和CYP2C9的表达产生显著的抑制效应(P<0.01)。结论黄芩苷对细胞中CYP3A4、CYP2C9和CYP2C19的表达具有影响作用,这种影响作用随药物处理时间不同而表现出双向效应。 【Objective】To find out the effects of Baicalin on CYP3A4, CYP2C9 and CYP2C19 expressions in Chang Liver cells. 【Methods】mRNA expressions of CYP3A4, CYP2C9 and CYP2C19 were determined by using RT-PCR in Chang Liver cells when incubated with DMSO 0.1% (v/v) (solvent control) and Baicalin (0.01 μM, 0.1 μM and 1 μM ) for 24, 36 and 48 hours, respectively. 【Results】CYP3A4, CYP2C9 and CYP2C19 expressions tended to increase when treated with different concentrations of baicalin in Chang Liver cells for 24 and 36 hours,respectively, and CYP3A4 and CYP2C9 expressions significantly decreased when incubated with different concentrations of baicalin for 48 hours (P 〈0.001). [ Conclusion] Baicalin had significant effects on CYP3A4, CYP2C19 and CYP2C9 expressions in a biphasic manner of first induction and later inhibition, and the effects varied with different concentrations of baicalin and incubated periods.
