扶正抑癌方含药血清对结肠癌lovo细胞侵袭转移能力及TGF-β_1与TβRⅡ蛋白表达的影响 被引量:13

摘要 目的:观察扶正抑癌方含药血清对结肠癌lovo细胞侵袭转移能力及TGF-β_1和TβRⅡ蛋白表达的影响,以探讨扶正抑癌方降低结肠癌转移的机制。方法:lovo细胞体外培养,制备大鼠不同浓度含药血清后,MTT测定不同浓度含药血清对细胞增殖的影响,以5-Fu作为阳性对照药;通过细胞迁移实验检测细胞运动能力,肿瘤细胞体外侵袭实验检测细胞侵袭能力,SABC免疫组化法检测TGF-β_1和TβRⅡ的蛋白表达。结果:高浓度含药血清24 h对lovo细胞作用有效,与对照组和空白组相比,扶正抑癌方含药血清能显著降低细胞的运动能力和侵袭能力,免疫组化提示扶正抑癌方含药血清能降低lovo细胞TGF-β_1的蛋白表达和提高TβRⅡ的蛋白表达,差异具有统计学意义(P<0.01)。结论:扶正抑癌方含药血清能有效抑制lovo细胞的增值和降低其侵袭力和运动能力,初步认为其作用机制与降低lovo细胞TGF-β_1的表达和升高βRⅡ的蛋白表达有一定的相关性。 Objective: To investigate the effects of drug serum of "Fuzheng Yi' ai Decoction" on invasion and metastasis capacity of human colon cancer cell line Lovo and its protein expression of TGF-β1 and TβRⅡ so as to probe the mechanism for the inhibition of the Decoction to metastasis of colon cancer. Methods: The Lovo cellswas cultured in vitro and the drug serum of different concentrations were prepared. The influence of drug serum on cell proliferation was detected by assay with 5-Fu as a positive control drug. The effects of drug serum of "Fuzheng Yi' ai Decoction" on migration and invasive capacity of lovo cells were detected by cell migration assay and matrigel invasion assay, respectively. The protein expression of TGF-β1 and TβR Ⅱ were detected by SABC immunohistochemistry. Results: After 24 hours, the drug serum of high concentration showed effects on the Lovo cells. Compared with the control group and blank group, the migration and invasive capacity of the cells in the drug serum group were significantly inhibited (P 〈0.01 ). The immunohistochemistry assay showed that the drug serum could reduce the protein expression of TGF-β1 and raise the expression of TβRⅡ, with a statistical significance (P 〈 0.01 ) . Conclusion: Drug serum of "Fuzheng Yi' ai Decoction" can inhibit the proliferation of lovo cells and reduce the cells' migration and invasive capacity, which may relate to decreasing the protein expression of TGF-β1 and increasing TβRⅡ.
