Rho激酶抑制剂Y-27632对人气道平滑肌细胞增殖和细胞周期的影响 被引量:3

摘要 目的观察Rho激酶抑制剂Y-27632对人气道平滑肌细胞(ASMCs)增殖的影响,为哮喘防治提供新思路。方法组织块贴壁法培养人ASMCs,对数传代后随机分为4组,A组加入含10%FBS的DMEM培养基,B、C、D组分别加入浓度为0.4、2.0、10.0μmol/L的Y-27632+含10%FBS的DMEM培养基,非放射性细胞增殖(MTS)/吩嗪甲酯硫酸盐(PMS)法检测各组细胞增殖情况(490 nm处A值),流式细胞仪分析细胞周期。结果C、D组A值均显著低于A组(P<0.05、0.01);C、D组G0/G1期细胞比例显著高于A组,S、G2/M期细胞比例显著低于A组(P<0.05、0.01)。结论Rho激酶抑制剂Y-27632可抑制人ASMCs增殖,阻滞细胞周期于G0/G1期;本研究为哮喘防治提供了新的选择。 Objective To investigate the effects of Rho-kinase inhibitor Y-27632 on the proliferation of human airway smooth muscle eeUs ( ASMCs), so as to provide new ideas for the treatment of asthma. Methods The ASMCs were primarily cultured to log phase by explant method and then divided into four groups, group A were treated with 10% FBS + DMEM, group B , C, D were treated with Y-27632 at the eoncentration of 0.4,2.0,10.0 μmol/L respeetively+ 10% FBS + DMEM. MTS/PMS assay was used to investigate the proliferation of ASMCs( A Value at 490 nm). Flow cytometry was used to ana- lyze the cell eyele. Results The A Value of group C and D were signifieandy lower than that of group A ( P 〈 0.05, 0.01 ). The cell percentages of G0/G1 phase of group C and D were much higher than that of group A,while cell ratios in S and G2/M were lower than that of group A (P 〈 0.05,0. 01 ). Conclusions Rho kinase inhibitor Y-27632 earl inhibit the proliferation of ASMCs. This study provides new options for the treatment of asthma.
