石楠藤提取物防治急性出血性坏死性胰腺炎大鼠内毒素血症的实验研究

Effect of Shinanteng extract on endotoxemia in rats with acute hemorrhagic necrotizing pancreatits

目的：观察石楠藤提取物对急性出血性坏死性胰腺炎（ＡＨＮＰ）大鼠内毒素血症的防治作用。方法：１００只ＳＤ大鼠随机分为３组：假手术组、ＡＨＮＰ组和石楠藤提取物治疗组（治疗组）。治疗组大鼠在ＡＨＮＰ诱发后１０分钟经腹腔内注射１％石楠藤提取物５ｍｌ／ｋｇ；假手术组和ＡＨＮＰ组大鼠腹腔内均注射０．５ｍｌ／ｋｇ生理盐水。结果：治疗组与ＡＨＮＰ组比较，血清淀粉酶值明显下降〔术后１２小时值为（１９．２９±５．３８）ｋＵ／Ｌ比（２８．４７±４．１５）ｋＵ／Ｌ，Ｐ＜０．０１〕；血中血小板活化因子（ＰＡＦ）含量明显降低〔术后１小时（１．３０±０．３０）μｇ／Ｌ，术后６小时（２．００±０．３４）μｇ／Ｌ，术后１２小时（２．１０±０．３２）μｇ／Ｌ比（２．２０±０．２５）μｇ／Ｌ，（６．１０±０．４２）μｇ／Ｌ，（１０．１０±０．５３）μｇ／Ｌ，Ｐ均＜０．０１）〕；血浆内毒素含量明显下降〔６小时（６９．９０±７．２０）ＥＵ／Ｌ，１２小时（９１．１０±８．４０）ＥＵ／Ｌ比（９８．４０±６．９０）ＥＵ／Ｌ，（１５１．７０±９．１０）ＥＵ／Ｌ，Ｐ均＜０．０１〕；治疗组胰腺组织的病理性损害较ＡＨＮＰ组明显减轻。结论：应用富含ＰＡＦ受体拮抗活?

Objective:To evaluate the treatment effect of Shinanteng extract on endotoxemia in rats with acute hemorrhagic necrotizing pancreatitis (AHNP).Methods:The model of AHNP was induced by retrograde infusion of DCANa into pancreatic duct.One hundred SD rats were divided into 3 groups:sham operation group (SOG),AHNP group (APG),and treated group(SNG,peritoneal injection with 1% Shinanteng extract in a dose of 5 ml/kg).Results:In the treatment group,serum amylase value was significantly decreased compared to AHNP group SNG vs.APG:(19 29±5 38) kU/L vs.(28 47±4 15) kU/L, P <0 01).Similarly,blood levels of plateletactivating factor (PAF) and endotoxin were also markedly declined PAF:(1 30±0 30)μg/L,(2 00±0 34)μg/L,and (2 10±0 32)μg/L vs.(2 20±0 25)μg/L,(6 10±0 42)μg/L,and (10 10±0 53)μg/L at 1,6,and 12 hours respectively,all P <0 01;endotoxin:(69 90±7 20)EU/L and (91 10±8 40)EU/L vs.(98 40±6 90)EU/L and (151 70±9 10)EU/L at 6 and 12 hours respectively,both P <0 01 .In addition,the lesion of pancreas was obviously mitigated in SNG compared to in APG.Conclusions:Shinanteng extract,containing abundant PAFreceptor antagonist,has beneficial effect in treating endotoxemia associated with AHNP in rats.
