摘要
用传统CTAB法和改良CTAB法提取盾叶薯蓣样品的基因组DNA。结果表明:传统CTAB法与改良CTAB法提取的基因组DNA完整性都比较好,但传统CTAB法获得的DNA含有较多杂质,不能被限制性内切酶消化,改良CTAB法提取的基因组DNA杂质去除彻底,条带清晰,可以完全酶切,并能获得清晰的预扩增图谱和有丰富条带稳定的AFLP指纹,完全适合于盾叶薯蓣分子标记鉴定所需。
The traditional CTAB method and the improved CTAB method were compared to extract the DNA of Dioscorea zingiberensis to select better method for AFLP analysis. Results showed that DNA obtained by the two methods all had good integrality, but the DNA obtained by traditional CTAB method contained more impurities and could not be digested by the restriction endonuclease. While the DNA extracted by the improved CTAB method was pure and had clear strip,so,it could be completely digested. And the preamplification figures was clear and had abundent, stable polymorphie AFLP fingerprinting figures. Therefore the improved CTAB was adapted to extract the DNA for AFLP Amplification reaction of Dioscorea zingiberensis.
