摘要
目的研究缬沙坦对早期糖尿病大鼠肾脏病变的影响。方法单侧肾切除SD大鼠随机分为3组:对照组(NC)、糖尿病组(DM)、治疗组(DV)组。治疗组于注射STZ当天起给予缬沙坦50mg/(kg.d)灌胃,疗程为8周。第8周时,观测肾组织形态学及肾功能变化,并采用免疫组化方法观察肾脏组织巨噬细胞浸润情况。结果DM组大鼠尿蛋白持续上升;肌酐清除率(Ccr)下降,肾脏组织巨噬细胞浸润增加。DV组以上指标均明显改善,DV组与DM组两组之间差异有统计学意义。结论缬沙坦对糖尿病大鼠早期肾脏损害有保护作用,其机制可能部分与抑制肾组织巨噬细胞浸润有关。
Objective To explore the effect of valsartan on abnormalities of kidneys of the new-onset diabetic rats.Methods SD rat removed the right kidney were randomly divided into 3 groups:diabetic valsartan treatment group(DV),diabetic non-valsartan treatment group(DM),normal control group(NC).Two weeks later,they were intraperitoneally injected with STZ at 35 mg/kg body weight daily for 3 days to induce diabetes.Valsartan 50 mg/kg body weight was administered through lavage in valsartan treatment group.Samples were obtained in the 8th week after diabetic model successfully established.The samples were measured by biochemistry parameters of blood and urine,renal tissue pathologic and immunohistochemistry.Results The 24 h urine protein was increased,creatinine clearance was reduced,CD68+ macrophages was decreased.After treatment with valsartan,the 24 h urine protein was reduced(P0.05),creatinine clearance was increased(P0.05),CD68+ macrophages was decreased(P0.05).Conclusion Treatment with valsartan may be involved in protection on kidney of new-onset diabetic animals partly via decreasement of CD68+ macrophages.
