甲醛和苯并(a)芘联合作用对DNA损伤的体外研究

摘要 以豚鼠肺巨噬细胞为研究对象,研究了甲醛和苯并(a)芘(BaP)在LD50、DNA-蛋白质交链(DPC)、DNA单链断裂(SSB)等3项指标上的联合作用。结果表明,甲醛和苯并(a)芘对豚鼠肺巨噬细胞的LD50分别为240.4mg/L和13.8mg/L,联合使用的LD50为61.7mg/L(其中甲醛为55.8mg/L,BaP为5.88mg/L),呈增强作用。甲醛能引起DPC,BaP不能引起DPC,联合作用时甲醛引起的交链程度降低;甲醛和BaP都能引起SSB,联合作用时SSB的程度增强。由此可见,甲醛和BaP对细胞杀伤和SSB具有增毒作用。 Alveolar macrophages of guinea pig were used in this study to investigate the effect of formaldehyde(HCHO) and benzo(a)pyrene (BaP) combination on LD50,DNA protein crosslinks (DPC) and DNA single strand breaks (SSB) in vitro.The results showed that LD50 of HCHO was 240 4mg/L,that of BaP was 13 8mg/L,and the combined LD50 was 61 7mg/L(HCHO was 55 82mg/L,BaP was 5 58mg/L).The type of combination was synergetic action.Only HCHO could induce DPC,and the intense of DPC decreased when used with BaP together.Both HCHO and BaP could induce SSB,and this effect increased when the two chemicals used together.So the combination on SSB was synergetic action too.\ \
