摘要
目的探讨行气活血、祛瘀生肌中药愈溃膏促进体表溃疡的愈合机制。方法用SPF级Wistar大鼠,参照塑料环肉芽肿定量法造模,随机分为单纯疾病模型组、生肌愈皮膏组、愈溃膏小、中、大剂量组,分别涂药干预,免疫组化法检测肉芽肿组织中VEGF、bFGF的表达。结果疾病模型组VEGF表达阳性率较低,经药物干预后,能够显著升高VEGF的表达(P<0.01,P<0.05),愈溃膏中、大剂量组效果最好;经药物干预后,bFGF的表达较单纯疾病模型组升高(P<0.01,P<0.05),以愈溃膏各剂量组作用明显(P<0.01),各剂量组间比较无显著性差异(P>0.05)。结论愈溃膏促进体表溃疡的愈合机制是通过增强肉芽肿组织中VEGF、bFGF的表达,增强血管生成,加速肉芽组织生长达到修复溃疡的目的。
Objective To explore the mechanism in promoting healing of body surface ulceration By Chinese herbal yukui ointment that acts on regulating qi,activating blood circulation,removing stasis and promoting the regeneration of muscle. Methods SPF Wistar rats were modeled in reference to plastic ring granuloma quantitative method and randomized into single disease model group and the groups of yukui ointment at small,middle and large dose. The ointment was applied for intervention. IHC method was adopted to determine the expressions of VEGF and bFGF in granuloma tissue. Results Positive expression rate of VEGF in disease model group was quite low and it increased significantly after drug intervention ( P 〈 0. 01 , P 〈 0.05 ). The efficacy in the groups of middle and large doses was the best. After drug intervention, bFGF expression increased compared with that in simple disease model group ( P 〈 0. 01, P 〈 0.05 ). The action of each dose group was obvious (P 〈 0.01 ). No significant difference presented in the comparison among groups of different doses( P 〉 0.05 ). Conclusion Yukui ointment promotes the healing of body surface ulceration by enhancing the expressions of VEGF and bFGF in granuloma tissue and angiogenesis, and accelerating the growth of granulation tissue so that ulcers can be repaired.
