内质网应激与胰岛素抵抗发生机制的关系

Relationship between endoplasmic reticulum stress and pathophysiology of insulin resistance

摘要 胰岛素抵抗(IR)与多种疾病密切相关,IR相关疾病的患病率逐年升高,近年来对于IR发生机制的研究成为热点。内质网应激(ERS)在IR的发生、发展中发挥重要作用,但其具体机制尚未完全明确。核因子κB(NF—κB)信号通路的激活与IR的发生、发展密切相关,并且与ERS间存在一定的联系,ERS可能通过激活NF—κB信号通路引起IR。 Recently many researchers focus on the pathogenesis of insulin resistance(IR) due to the gradually increased prevalence of diseases which are closely related with IR. Accumulating evidence suggests that endoplasmic retieulum stress(ERS) plays a role in the pathogenesis of IR, but the mechanism has not been clarified clearly. The nuclear factor kappa B(NF-κB) pathway is activated in various tissues under IR condition,and suppresssion of NF-κB pathway can improve IR. It has been proved that ERS leads to activation of NF-κB pathway. Thus,NF-κB pathway plays an important role in pathogenesis of IR and may be a connection between ERS and IR.
