摘要
为系统观察脾虚和肾虚雌性SD大鼠的生育能力,将大鼠随机分为:脾虚组、脾虚复健组、肾虚组、肾虚复健组、正常组。脾虚组和肾虚组分别按常规方法用利血平和羟基脲制成脾虚模型和肾虚模型。虚证动物模型出现与人类相似的症状:动情周期紊乱,甚至消失;生殖器官萎缩;血清卵泡剌激素(FSH)、雌二醇(E2)水平下降;子宫腺体数目减少,卵巢次级和成熟卵泡数减少,生育能力下降。而肾虚大鼠上述变化更显著。用四君子汤喂饲脾虚模型和用肾气汤喂饲肾虚模型后,大鼠脾虚、肾虚症状得到明显改善,肾虚复健组大鼠生育能力提高更显著。结果表明脾虚直接影响消化系统功能,间接影响内分泌功能(包括下丘脑-垂体-性腺轴,即HHS轴功能),而肾虚直接影响HHS轴功能;四君子汤和肾气汤都有补虚作用,能调整HHS轴功能。为中医“脾虚及肾”、“肾主生殖”
Genitality in female SD rats with spleen deficiency and kidney deficiency was observed systematically. The spleen-deficiency rat(SDR) model was set up with injection of reserpine and kidney_deficiency rat (KDR) model by gastric infusion of hydroxycarbamide. Si Junzi Tang and Shenqi Tang were used to recover the SDR and KDR respectively. Rats were allocated to five groups: SDR group, Si Junzi Tang SDR group, KDR group,Shenqi Tang KDR group and normal control group. It was found that results in the rats with deficiency syndromes were similar to those in human being,for instance, sexual cycle became irregular or even disappeared; genital organs went atrophy; the level of serum FSH and estradiol decreased; the numbers of uterine glands, secondary follicle and mature follicle lowered and the genitality decreased. The above changes occurred in the KDR especially. Symptoms in the deficiency syndromes were improved by invigorating the spleen and tonifying the kidney. The genitality was obviously increased in Shenqi Tang KDR group. It is indicated that in SDR the digestive function is affected directly and the endocrine function indirectly (including hypothalamus_hypophysis_gonad axis,i.e.,HHG axis), while in KDR, HHG axis is affected directly. Both Si Junzi Tang and Shenqi Tang can regulate HHG axis. The theory of deficiency spleen involving the kidney and the kidney controlling the genitality in TCM was also discussed.
