摘要
目的:探讨锯叶棕提取物与AG490合并应用后对MM细胞增殖的影响。方法:将体外培养的U266和PRM182:26细胞与0—2uL/mL的不同浓度锯叶棕提取物共孵育,应用MTT比色法测定细胞增殖活性;同时应用台盼蓝不相容实验检测锯叶棕提取物与AG490单独使用或合并使用时的细胞成活率。结果:锯叶棕提取物抑制U266与RPM18226细胞的增殖,其抑制生长50%(ED50S)的有效剂量分别为1.2和0.7灿L/mL;U266细胞用锯叶棕提取物(0.5uL/mL)或AG490(20nM)预处理,其细胞成活率分别为(79.7±1.1)%和(67.2±6.1)%,两者联合应用细胞成活率为(42.9±12.0)%(P〈0.001);RP—M18226细胞用锯叶棕提取物(0.5ul/mL)或AG490(20nM)预处理,其细胞成活率分别为(74.7±2.1)%和(37.2±5.1)%,两者联合应用细胞成活率为(42.9±4.1)%(P〈0.001)。结论:锯叶棕提取物与AG490合并应用抑制MM细胞的增殖作用较单独使用效果好。
Objective: To investigate the influence of extract of Sababae serrulatae combined with AGd90 on cell proliferation of multiple myeloma. Method : U266 and PRMI8226 cells cultured in vitro with extract of Sababae serrulatae 0-2 uL/mL in different concentrations were incubaied together. The activity of cell proliferation was assayed using MTT colorimetry. And at the same time trypan blue exclusion test detected the cell survival rate of extract of Sababae serrulatae used alone or in combination with AG490. Result : Extract of Sababae serrulatae inhibited the proliferation of U266 and RPMI8226 cells, and the effective doses which could inhibit 50% growth of (EDSOS) were 1.2 and 0.7 uL/mL respectively. After pretreatment of U266 cells by extract of Sababae serrulatae (0.5 uL/mL) or AG490 (20nM) ,the cell survival rates were 79.7± 1.1% and 67.2 ±6.1%. And the celt survival rate of combined pretreatment by both of them was 42.9± 12.0% (P 〈 0. 001 ). And after pretreatment of RPMI8226 cells by extract of Sababae serrulatae (0.5 uL/mL) or AG490 (20 riM) ,the cell survival rates were 74.7 ± 2.1% and 37.2 ± 5.1%. But the cell survival rate of combined pretreatment by both of them was 42.9± 4.1% ( P 〈 0.001 ). Conclusion : Extract of Sababae serrulatae combined with AG490 can inhibit cell proliferation of MM. And combination application has better effect than alone using.
