金欣口服液含药血清对呼吸道合胞病毒感染细胞早期凋亡的影响 被引量:18

摘要 目的:探讨金欣口服液含药血清对呼吸道合胞病毒(RSV)感染细胞早期不同时点凋亡及Bcl-2、Bax表达的影响。方法:采用细胞培养、血清药理学、流式细胞术等实验技术检测金欣口服液含药血清抗RSV,影响细胞早期凋亡作用途径的影响。结果:RSV感染人胚肺成纤维细胞(HELF)细胞12、24h后凋亡率均低于细胞对照组(P<0.05),Bcl-2/Bax比例逐步上调,金欣口服液作用后,凋亡率明显高于病毒组(P<0.05),Bcl-2/Bax比值明显低于病毒组(P<0.05)。结论:金欣口服液在RSV感染早期,可能通过调控凋亡相关基因拮抗其抑制细胞凋亡的作用,从而影响进入细胞内的病毒复制增殖的过程,阻止病毒的扩散。 Objective: To study the effect of Jinxin Oral Liqiud medicated serum on RS virus virus (RSV) infected cell apoptosis and the regulation gene as Bcl-2, Bax on the different point in early time. Methods: Cell culture, serum pharmacology and Annexin V/PI technique combining with flow cytometry were used to detect cell apoptosis, Bcl-2 and Bax expression. Results: The total apoptosis rates of human embryonic lung fibroblast in JOL group in early time of 12h and 24h were significantly higher than that in RSV infected group (P〈0.05). The expression of Bcl-2/Bax in JOL group was lower than that in virus group (P〈0.05). Conclusion: The mechanism of JOL inhibited RSV maybe relate to affecting the early apoptosis of the RSV-infected cells through suppressing the Bcl-2 and uprising the Bax expression, which reduced the RSV diffusion.
