摘要
目的:观察急性胰腺炎(AP)大鼠肺组织中信号转导与转录激活因子-1(STAT-1)及肿瘤坏死因子-α(TNF-α)的表达,探讨磷酸化STAT-1(P-STAT-1)在AP肺损伤中的作用。方法:40只Wistar大鼠随机分为假手术组(CON组)10只;AP模型组(AP组)30只,其中3、6、12h时间亚组各10只。CON组仅翻动胰腺数次后关腹,AP组以约0.1ml/min恒速逆行注射5%牛磺胆酸钠制备AP模型。术后3、6、12h分批处死大鼠收集标本,测定各组血清淀粉酶(AMY),聚合酶链反应(PCR)检测肺组织TNF-αmRNA表达,Western-blot检测肺组织P-STAT-1蛋白表达。结果:AP组各时间点AMY、TNF-αmR-NA和P-STAT-1蛋白的表达明显高于CON组(P<0.05)。AP组P-STAT-1蛋白在3h表达升高,6h后达到高峰,12h明显下降。AP组TNF-αmRNA的表达3h开始增加,12h达到高峰。结论:AP大鼠肺组织中STAT-1蛋白的活化可能影响TNF-α等炎症因子的表达,并在AP合并肺损伤的发病过程中起一定作用。
Objective:To observe the expression of STAT-1 and TNF-α in lung tissue of rat with severe acute pancreatitis(AP),to explore effect of P-STAT-1 on the lung injury of rat with AP.Method:40 Wistar rats randomly divided into two group:sham operating group(10 rats)and AP group(30 rats)including different time points in 3,6,12h after the operation,each point have 10 rats.CON group:sham-operated animals served as control.AP group:AP was induced by retrograde injection of 5%sodium deoxycholate(1ml/kg)into the biliopancreatic duc.3h,6h,and 12h after the operation,rat was executed and the sample was collected,serum amylase,expression of P-STAT-1 protein and TNF-α mRNA in lung tissue were measured.Results:Compared to the CON group,the serum amylase,expression of P-STAT-1 protein and TNF-α mRNA in the lungtissue elevated every time point in AP group(P<0.05).In AP group,expression of P-STAT-1 protein,increased at 3h after the operation,reached its peak at 6h and decreased obviously at 12h after the operation.The expression of TNF-α mRNA increased at 3h and reached its peak at 12h after the operation.Conclusion:Activation of STAT-1 protein in lung tissue of rat with AP can affect the expression of the inflammatory factor like TNF-α,which may play an important role in pathogenesis of lung injury complicated by AP.
