The effect of IL-2 in mouse model with vaginal candidiasis

摘要 目的:通过对小鼠阴道白假丝酵母菌病感染过程中IL-2的变化,探讨其在阴道白假丝酵母菌病细胞免疫中的作用。方法:将试验小鼠分为3组,分别为一次感染组(A组)、二次感染组(B组)、对照组(C组),通过小鼠阴道种植白假丝酵母菌制做小鼠阴道白假丝酵母菌感染模型,分别于2、7、14天取小鼠阴道灌洗液及血液测IL-2的含量,并留取部分阴道灌洗液做真菌培养,观察两者之间的联系。结果:A组与B组血液及阴道灌洗液中IL-2含量在感染第7天达到高峰,感染第2天及第14天含量无显著变化,含量均高于对照组。B组同时期IL-2含量较A组显著增多。A组及B组同时期阴道灌洗液中IL-2含量较血液中显著增多。阴道灌洗液真菌培养显示A、B组CFU计数于第7天达到高峰,第14天与第2天无显著差异。B组同时期真菌培养CFU计数比A组显著增多。结论:IL-2在抗白假丝酵母菌的感染过程中发挥了作用,主要是通过阴道局部粘膜免疫发挥作用,且在再次感染过程中发挥较大作用。 Objective: To study the effect of IL -2 in mouse model with vaginal candidiasisMethods: 63 female Kunming mice were divided into three groups. Group A was infected by Candida alb cans only once, Group B was infected two times and group C was the control group, IL-2 in blood and vaginal fluids were detected and CFU of vaginal fluids was performed after infection of 2, 7 and 14 days. The results were processed by SPSS 13.0 statistical software. Results: The content variation of IL -2 and CFU in vaginal fluids was the highest at the 7th. day in both group A and group B, as well as in the blood; there were not notable changes at 2nd. day and 14th. day. The content of IL - 2 in group A and group B were all higher than group C. According to the same days post - infection, IL - 2 of the group B was higher than that of group A both in vaginal fluids and in blood, and the IL - 2 in vaginal fluids was higher in blood. The CFU of group B was higher than group A. Conclusion: Cell - mediated immunity ( CMI), as a host defense mechanism, plays an important role in protecting against Candida albicans, especially in secondary infection. Local innate immunity is more important than systemic innate immunity in vulvovaginal candidiasis. IL- 2 has protective affection in VVC by promoting local innate immunity level.
