摘要 目的探索建立柔红霉素(DNR)血管外渗动物模型的最佳药物外渗剂量。方法使用5种不同剂量的柔红霉素皮下注射家兔局部皮肤,观察局部皮肤损害和病理学特征。结果A组(生理盐水对照组)皮肤无明显异常;B组DNR 0.2 mg/mL(与人体使用剂量相同)没有引起家兔局部组织坏死;C、D、E组(DNR 1、2、3mg/mL)均可引起内皮细胞脱落、炎性细胞浸润、组织水肿及不同程度溃疡和坏死,3组之间差异无统计学意义P>0.05);F组(DNR 4 mg/mL)可致局部组织出现明显溃疡和坏死。结论采用1、2、3 mg/mL的DNR均可建立致家兔柔红霉素血管外渗的动物模型,为提高实验研究的稳定性,建议2 mg/mL为最佳药物外渗剂量。 Objective To investigate the best dosage of daunorubicin to establish a daunorubicin extravasation animal model. Methods Intradermal daunorubicin injection of 5 different dosages in New Zealand rabbits provided re- producible skin lesion, which were observed and examined under microscope. Results Control group (Group A.. NS) revealed no lesion;Group B (DNR 0.2 mg/mL, equal to the dosage in human being) revealed no lesion;Group C, D, E (DNR 1 mg/mL, 2 mg/mL, 3 mg/mL, respectively)revealed endothelia shedding, inflammatory cells infiltration, epider- mis edema, ulceration and necrosis in varying degrees. No significant difference ( P 〉 O. 05 ) there was within the 3 groups;Group F (DNR 4 mg/mL) led to obvious ulceration and necrosis. Conclusion Intradermal daunorubicin injec- tion of 1,2 and 3 mg/mL provide reproducible skin lesion in rabbits, which provided animal model of daunorubcin extravasation. Two mg/ml was the recommended dosage.
