The effects of bone marrow mesenchymal stem cell transplantation on angiogenesis in neonate hypoxic-ischemia brain damage rats

摘要 目的分离培养鼠骨髓间充质干细胞(BMSCs)通过脑室注射移植给缺血缺氧性脑损伤(HIBD)大鼠,观察其对大鼠影响。方法从97只SD大鼠随机选取66只进行HIBD造模(HIBD组34只,治疗组32只),余31只作为对照组。治疗组予脑室注射BMSCs,对照组及HIBD组予脑室注射等量生理盐水,分别于移植1、3、7、14d处死各组大鼠,免疫组织化学法检测Brdu阳性细胞和VEGF表达。结果移植后第3、7、14天均从治疗组大鼠脑组织切片检出Brdu阳性细胞,而HIBD组各时点均未检出Brdu阳性细胞。而移植后第一天,HIBD组和治疗组脑组织中均有VEGF表达,但强度差异无显著性;第3、7天,治疗组表达强于HIBD组,差异有显著性;第14天两组表达强度均减弱;而对照组各时点表达均较弱。结论BMSCs通过脑室内注射的进入脑组织成活并定植,可增强VEGF表达。外源性的细胞移植能促进损伤脑组织血管再生,从而减轻脑组织缺氧缺血损伤的程度。 Objective To explore the angiogenesis effect of BMSCs from human umibilical cord blood after ransplantation into cerebral ventricle in HIBD rats. Methods Neonate HIBD rats were randomized into experimental group and control group. The BMSCs marked with BrDU were injected into the cerebral ventricle in experimental group, with the control group received saline solution. Subsequently, immunohistochemistry was applied to assess angiogenesis. Results The expression of VEGF in experimental group was significantly higher than that in the control group on day 1, 3, 7 and 14 postoperatively. There was no significant difference in VEGF expression between the treatment group and thecontrol group. However,VEGF expression was significantly higher in experimental groups than that in control group on day 3 and 7, but returned to normal on day 14. BrDU positive cells were detected in experimental rat brains on day 14. Conclusion BMSCs injected into the cerebral ventricles of HIBD can replant in cerelral tissue and up - regulate VEGF expression. Thus, transplantation of BMSCs induces angiogenesis and reduces HIBD.
