摘要
目的旨在探讨12-脂氧化酶(12-LOX)抑制剂黄芩素(Baicalein,BAI)对人食管癌EC109细胞增殖及细胞周期的影响及可能的机制。方法不同浓度BAI(0、5、10、20、40、80μmol/L)处理EC109细胞,应用MTT法检测BAI对EC109细胞增殖的影响;流式细胞术(FCM)检测BAI对EC109细胞周期的影响。结果随着BAI终浓度(0、5、10、20、40、80μmol/L)增加和作用时间(24、48、72 h)的延长,细胞存活率明显下降;24 h后分别为98.4%、87.7%、87.6%、81.9%、79.0%;48 h后分别为71.6%、67.6%、64.1%、60.0%、46.7%;72 h后分别为43.8%、43.1%、43.1%、40.0%、23.0%;FCM分析表明:药物作用24 h后,随着BAI浓度的增高,G1期细胞的比例由43.6%升高至68.5%,G2期细胞的比例从24.5%降至0.0%。结论BAI能抑制EC109细胞的增殖且改变细胞周期的分布,并呈一定的时间、浓度依赖性。
Objective To investigate the effects and possible mechanisms of Baicalein on the proliferation and cell cycles of human esophageal carcinoma cell line EC109. Methods EC109 ceils were treated with different BAI concentration(0, 5, 10, 20, 40 and 80(mol/L). Thiazolyl blue terazolium bromide (MTT) method and flow cytometry (FCM) were used to measure cell proliferation and cell cycle of EC109 cells, respetively. Results With the concentra- tion from 0 to 80( mol/L and exposure time form 24 to 72 hours, BAI inhibited the proliferation of EC109 cells in concentration - dependent and time - dependent manners. Rating by concentration from 0 to 80( mol/L, the survival rates of cells were 98.4%,87.7%,87.6%,81.9% and 79.0%, respectively, in 24 h, 71.6%,67.6%,64.1%,60.0% and 46. 7%, respectively, in 48h and 43.8% ,43. 1% ,43.1% ,40.0% ,23.0% ,respectively,in 72 h. Cell cycle analysis revealed a marked increase from 43.6% to 68. 5% in the G1 phase and a decrease from 24. 5% to 0. 0% in the G2 phase following 24 h exposure to BAI in a concentration - dependent manner. Conclusion BAI inhibits the proliferation of ECI09 cells in concentration - dependent and time - dependent manners. BAI may change the cell cycles in a concentration - dependent manner.
